dc.description.abstract | Der vaskuläre endotheliale Wachstumsfaktor (VEGF) ist ein Schlüssel-Mediator der Angiogenese. Bei Sauerstoffmangel (Hypoxie) wird die VEGF-Expression durch die HIF-1-vermittelte Transkription, die Stabilisierung der VEGF-mRNA und die IRES (interne Ribosomen-Eintritt-Stelle)-vermittelte Translation verstärkt. Zwei IRESs in der 5’-untranslatierten Region der VEGF-mRNA erhalten deren Translation unter Hypoxie aufrecht, während die 5‘-Cap-abhängige Translations-Initiation inhibiert wird. Zur Identifizierung von in trans-agierenden Faktoren wurde, basierend auf normoxischen und hypoxischen MCF-7-Zell-Extrakten, eine Tobramycin-RNA-Affinitäts-Chromatographie durchgeführt. Die DEADbox RNA-Helikase 6 (DDX6) konnte als VEGF-mRNA-interagierendes Protein identifiziert werden. Rekombinantes DDX6 inhibiert die VEGF-IRES-vermittelte Translation in normoxischen MCF-7-Zellextrakten in vitro, während die Depletion von DDX6 mittels RNAi die VEGF-Expression in hypoxischen MCF-7-Zellen stimuliert. | - |
dc.description.abstract | The vascular endothelial growth factor (VEGF) is a key-mediator of angiogenesis in physiological and pathological conditions. Under low oxygen conditions (hypoxia) VEGF expression is strongly enhanced via its HIF-1 dependent transcription, VEGF mRNA stabilization and internal ribosome entry site (IRES)-mediated translation. Two independent IRESs in the VEGF mRNA 5’-untranslated region confer sustained VEGF mRNA translation under hypoxia when 5'-cap-dependent mRNA translation is reduced. VEGF IRES-mediated initiation of translation requires the modulated interaction of trans-acting factors. An RNA affinity purification approach based on extracts derived from MCF-7 cells cultured under normoxia or hypoxia (1% oxygen) revealed the DEAD-box RNA helicase 6 (DDX6) as VEGF mRNA trans-acting factor. Recombinant DDX6 inhibits VEGF IRES-mediated translation in normoxic MCF-7 cell extracts in vitro while depletion of DDX6 by RNAi promotes VEGF expression in hypoxic MCF-7 cells. | eng |
